Title: Antidepressant-like effect of essential oil of cymbopogon flexuosus in a chronic unpredictable mild stress-induced depression model in mice

Abstract: Cymbopogon flexuosus, a famous Indian perennial grass containing citral, is traditionally used to alleviate stress, anxiety and depression. Cymbopogon flexuosus, commonly known as lemongrass-are cultivated mainly for essential oils and are native to India. Due to lack of experimental studies, it is prerequisite to evaluate the antidepressant potential of C. flexuosus in experimental animals. The present study was employed to explore the antidepressant potential of C. flexuosus in chronic unpredictable mild stress (CUMS) model induced depression-like behaviour in mice and to identify probable mechanism of action of C. flexuosus in amelioration of oxidative stress and depression. Mice were subjected to 28 days of chronic unpredictable mild stress (CUMS) procedure, and treated with Cymbopogon flexuosus (50 and 100 mg/kg, po) or imipramine (15 mg/kg, po). CUMS induced depression like behavior was assessed by forced swimming test (FST) and tail suspension test (TST) and followed by treatment with Cymbopogon flexuosus. Furthermore, oxidative stress parameters were determined by measuring the levels of reduced glutathione (GSH), malondialdehyde (MDA) formation, nitrite, superoxide dismutase (SOD), catalase (CAT). Lemongrass oil or imipramine significantly decreased immobility in CUMS mice as evaluated by FST and TST. In addition to this, lemongrass oil decreased malondialdehyde formation and nitrite levels, while glutathione, superoxide dismutase and catalase levels were found to be increased in the whole brain. The results of the present study demonstrated that lemongrass oil attenuated the CUMS-induced depression in mice, by reduction of oxidative´┐Żnitrosative stress in the mice brain.
